"A. FEATURE
The Rotor-GeneTM is the only real-time analyzer to employ a centrifugal rotary design. There are many benefits to the rotary format including better performance, versatility, robustness and easier maintenance.
1. Thermal Uniformity
The Rotor-GeneTM tubes are arranged in a circular rotor that spins continuously at 400 rpm in a chamber of moving air. Thus there is no temperature variation between tubes due to positional effects such as the recognized “edge effect” observed in block-based designs.
2. Optical Uniformity
Optically, the Rotor-GeneTM is similarly uniform because every tube moves past the identical excitation light source and detection pathway. Optical calibration is a topic few vendors like to discuss because elaborate optical alignment and calibration is usually required. In the Rotor-GeneTM this isn’t needed because the identical optical path reaches every sample. Furthermore, the Rotor-GeneTM doesn’t require a normalization dye (like the the ROX™ passive reference) for the same reason.
3. Light-source Guarantee
The Rotor-GeneTM uses a separate color high-power light-emitting diode (LED) as an excitation source for each channel. LEDs maintain a uniform output over time and we provide a lifetime replacement guarantee.
4. High-Speed Data Acquisition
All tubes in a rotor pass the detector every revolution (150 milliseconds), allowing data to be collected at a fast rate. New and demanding applications such as HRM (high-resolution melt) rely on fast data acquisition and can thus be accommodated.
5. Analysis Options
The Rotor-GeneTM software gives you more real-time data analysis options as standard, including many you won’t find elsewhere. For example, choose from several methods for gene expression and quantification analysis, including REST (relative expression software tool), delta-delta CT (comparative CT method), LinReg (Linear Regression/Assumption-Free Analysis), Comparative Quantification, Standard Curve Quantification, 2 Standard Curves Relative Quantification. In addition, you can determine the amplification efficiency of individual reactions or export the raw data for external analysis.
6. Concentration Analysis
The Rotor-GeneTM is fully equipped to do concentration analysis from fluorescence measurements. A typical application is the measurement of DNA concentration using PicoGreen dye (see example).
7. Optical Temperature Verification
More and more laboratories now require regular verification testing of instrument performance as part of their standard operating procedures. This is especially true for laboratories with validated protocols that must remain repeatable and transferable.
